目的 探讨 DOPEY2在 TMPRSS2-ERG基因融合阴性前列腺癌中的表达及其临床意义。方法 本研究通过 RT-PCR和 Western Blot技术检测前列腺癌细胞株 VCaP和 PC-3中 ERG和 DOPEY2的表达情况,同时,采用免疫组化技术检测 46例前列腺癌根治术后的石蜡标本中 ERG和 DOPEY2的表达情况,分析两者的表达以及与临床病理特征之间的关系。结果 DOPEY2在 TMPRSS2-ERG基因融合阳性前列腺癌细胞株 VCaP中的 mRNA和蛋白表达水平,均比 TMPRSS2­ERG基因融合阴性前列腺癌细胞株 PC-3低,且差异均有统计学意义( P<0.05)。免疫组化结果显示, DOPEY2在 TMPRSS2-ERG基因融合阴性组织中的阳性表达率高于基因融合阳性组( 76.5%和
Objective To study the difference and clinical significance of DOPEY2 expression between TMPRSS2-ERG gene fusion-positive and TMPRSS2-ERG gene fusion-negative prostate cancer. Methods The expressions of ERG and DOPEY2 in two prostate cancer cell lines (VCaP and PC-3) were examined by RT-PCR and Western Blot. Then, immunohistochemistry was used to detect the expression of ERG and DOPEY2 in 46 cases of prostate cancer. Results The mRNA and protein expression levels of DOPEY2 in TMPRSS2-ERG gene fusion-negative cell line (PC-3) were significantly higher than that in TMPRSS2-ERG gene fusion-positive cell line (VCaP, P<0.05). The results of immunohistochemistry showed that the expression ratio of DOPEY2 in ERG-negative and ERG-positive prostate cancer was 76.5% and 33.3% respectively (P<0.05). In addition, DOPEY2 was positive correlated with Gleason Grade and T stage (P<0.05). Over-expression ratio of ERG protein was 26.1% (12/46), and it had significant correlation with low Gleason score (P<0.05), while not with age, serum PSA levels, T stage and metastasis. Conclusion s There is a significant inverse correlation between the expression of ERG and DOPEY2 in both prostate cell lines and cancer tissues. DOPEY2 can serve as a helpful and prognostic biomarker in prostatic cancer.
