摘要:
目的建立可应用于活体成像的原位膀胱癌动物模型并利用生物发光体外活体成像系统检测肿瘤的生长。方法通过感染能表达荧光素酶的重组慢病毒颗粒,将荧光素酶基因转染至膀胱癌细胞 BIU87中。然后向 6只裸鼠膀胱壁内注射肿瘤细胞 BIU87-luc,构建原位膀胱癌模型,并在肿瘤细胞种植后的第 5天及第 10天,利用生物发光体外活体成像系统检测肿瘤的变化。最后取各个裸鼠的膀胱制作病理切片观察。结果 BIU87与 BIU87-luc两种细胞的生长、迁移能力没有明显差别。膀胱壁内注射肿瘤细胞后第 5天,所检测到的光子信号强度为( 1.78±0.48)×106 p/sec/cm2/sr,第 10天所检测到的光子信号强度为( 5.07±0.81)×106 p/sec/cm2/sr,二者的差别有统计学意义( P<0.01)。最终的膀胱病理切片可以看到大量肿瘤细胞聚集生长。结论荧光素酶标记膀胱癌细胞后,构建的原位膀胱癌动物模型,利用活体成像技术能够动态监测其原位肿瘤的生长。
